dc.description.abstractEl virus del Herpes simple tipo 1 (VHS-1) infecta a humanos causando lesiones epiteliales leves en la mayoría de los infectados, sin embargo, en pacientes inmunocomprometidos se podrían desarrollar casos graves como encefelatis herpética con compromiso de la vida. Se ha observado una mayor frecuencia de aislamiento de cepas resistentes al tratamiento antiviral. Debido a ello es necesario el desarrollo de nuevas fuentes de tratamiento, las especies vegetales constituyen fuentes interesantes para la búsqueda de nuevos fármacos. En este proyecto se evaluó la actividad antiviral de 28 extractos metanólicos de plantas del Paraguay mediante qPCR. Se determinó que 10 extractos presentaron actividad antiviral asociada a una baja citotoxicidad. Además, se analizaron algunas fracciones, donde se destacan por su actividad antiviral las fracciones hexánicas y cloróformicas de Acanthospermum australe y A. hispidum. Se observó un IS de 49,35 y 430,58 para las fracciones clorofórmicas de A. australe y A. hispidum, y de 12,38 y 521,41 para las fracciones hexánicas de A. australe y A. hispidum. La evaluación del mecanismo de acción permitió determinar que las fracciones hexánicas actúan a nivel celular en la etapa de pre y pos adsorción. Mientras que las fracciones clorofórmicas de ambas especies actúan en todas las etapas del ciclo viral. En cuanto a la composición química de las fracciones activas se determinó mediante LC-MS la presencia de 5,7,3´,4´-tetrahidroxi-3,6-dimetoxiflavona o axilarina, 5,7,4´trihidroxy-3,6-dimetoxiflavona o 3,6 dimetoxiapigenina, 5, 4' dihidroxi 3,6,7 trimetoxi flavona o penduletina, 3',4',5-trihidroxi-3,6,7-trimetoxiflavona o crisosplenol D, (Z,Z)-9,12-octadecadienoato de metilo, y fitol. Estos compuestos podrían ser responsables de la actividad antiviral, sin embargo, no se descarta que pueda haber otros compuestos que contribuyan a la misma y que no han podido aún ser identificados. Además, demuestra que el método de qPCR constituye una herramienta efectiva para la identificación de productos vegetales con actividad antiviral.es
dc.description.abstractThe Herpes simplex virus type 1 (HSV-1) infects humans causing mild epithelial lesions in the majority of those infected, however, however, in immunocompromised patients, severe cases such as herpetic encephelatis with life compromise could develop. A higher frequency of isolation of strains resistant to antiviral treatment has been observed. Because of this, it is necessary to develop new sources of treatment, plant species are interesting sources for the search for new drugs. In this project, the antiviral activity of 28 methanolic extracts from Paraguayan plants was evaluated using qPCR. It was determined that 10 extracts presented antiviral activity associated with low cytotoxicity. In addition, some fractions were analyzed, where the hexane and chloroform fractions of Acanthospermum australe and A. hispidum stand out for their antiviral activity. An IS of 49.35 and 430.58 was observed for the chloroform fractions of A. australe and A. hispidum, and of 12.38 and 521.41 for the hexane fractions of A. australe and A. hispidum. The evaluation of the mechanism of action made it possible to determine that the hexane fractions act at the cellular level in the pre and post adsorption stage. These compounds could be responsible for the antiviral activity; however, it is not ruled out that there may be other compounds that contribute to it and that they have not yet been identified. Furthermore, it shows that the qPCR method constitutes an effective tool for the identification of plant products with antiviral activity.en
